Test Utility

The test is a semiquantitative culture method that identifies up to two pathogenic bacterial organisms with concentrations greater than 10,000 cfu/mL.  Antibiotic susceptibility testing will also be performed, if indicated for the identified organism(s). Urine culture results should be used in conjunction with other tests to identify the causative organism(s) of urinary tract infections.

Specimen Collection and Preparation

Sterile Urine Cup / Boric Acid tube

  1. Do not centrifuge

  2. Urine collected in a sterile urine container:

  3. > 4 mL: Transfer to a Boric Acid tube (preserved urine) - see below for stability

  4. < 4 mL: Do not transfer to a Boric Acid tube (unpreserved urine) - see below for stability

Specimen Storage and Stability

  1. Preserved urine in Boric Acid tube (preferred):

  2. Store at room temperature (20-26°C) up to 24 hours

  3. Store refrigerated (2 to 8° C) up to 48 hrs

  4. Unpreserved urine (original collection container or < 4 mL volume)

  5. Store refrigerated (2 to 8° C) up to 24 hours


Note: Improper or delayed specimen handling can result in overgrowth of commensal (normal) flora resulting in inaccurate results. Organism growth will continue if the specimen is left at room temperature longer than 2 hours.

To reduce misleading or false positive results and improve patient care, urine specimens should ideally be transferred to a Boric Acid tube within 2 hours of collection and stored refrigerated up to 48 hours. Please follow the guidelines above and call the laboratory if you have any questions.

Specimen Rejection

  • Underfilled Boric Acid tubes (<4 mL)

  • Leaking or unsuitable specimen container

  • Insufficient sample volume

  • Specimen labeled incorrectly

  • Sample not stored properly

  • Sample is frozen

  • Sample received outside of stability

  • Specimen contamination – (i.e. lubricants, talc, contrast media, fecal material)
