Methodology

Direct (conjugated) bilirubin couples directly with a diazonium salt of 3,5-dichloroaniline (DPD) in an acid medium to form azobilirubin. The direct bilirubin in serum is directly proportional to the color development of azobilirubin which is measured biochromatically at 570/660 nm.

Test Utility

An indication of hepatic function and biliary disease, including hepatitis, cirrhosis, and other advanced neoplastic diseases involving the liver.

Specimen Collection and Preparation

Serum or plasma sample in the following tubes:

  • Serum separator tube (SST or tiger top)*

  • Lithium Heparin (green top)*

*Protect from direct light as soon as possible

Specimen Storage and Stability

Store serum or plasma at the following temperatures:

  • Room Temperature: Not indicated

  • Refrigerated: 2°C to 8°C up to 3 days*

  • Frozen: -20°C or lower up to 3 months*

*Protect sample from light

Specimen Rejection

  • Insufficient sample volume

  • Sample not stored at proper temperature

  • Sample that has been excessively exposed to light.

  • Frozen whole blood

  • Hemolyzed samples

  • Severely lipemic specimens

  • Specimen labeled incorrectly or not labeled
